I met my wife on line. my profile was something like "strong mind in strong body, looks like a fireplug in a suit" something like that. put down the 3 or 4 important points that you think people should know, and put in any negatives that you think will take you out of the running, no need to waste dates on girls who won't go for you for a reason that will be obvious once they see you.

I'd keep the profile short and positive, personally. I bring up "negatives" in first couple of emails. I hate clubbing/bars/etc and would always mention that in response the inevitable question about what I like to do for fun. It's easy to make yourself seem better than you really are online. Personally, I tried hard not to do that. Use pics that you look good in but that show what you really look like. I would tailor the rest of your profile towards the type of woman you want to attract. A profile targeted at successful, educated women that are looking to settle down is going to be very different than one targeted at university students imo.

Whatever you write, make it light hearted and humorous. It will be memorable and you won't turn people off for appearing to take yourself seriously. If that fails, start sending other members dollar signs.

But in all seriousness, I'd probably just write about a couple hobbies/activites that you genuinely enjoy and injecting your flavor of humor into it. I'd probably shy away from the Barney Stinson profile.
