My story mimics Dakota's. The guy was my best friend and I'd always gotten along with his younger sister. I asked him outright what he thought of idea and his response was similar to the effect of "it's really none of my business. You're dating her, not me." We dated for a few months and ended it amicably as we were both in different colleges.
Moral of the story is that my best friend is still my best friend and I still get along great with his sister when I see her (and she's even hotter now). I think it comes down to the kind of person your friend is with regards to getting permission to pursue his sister, but I think if I had to do it over again I'd come up with one hell of an exit strategy if things weren't working out unless I wanted to lose a friend. I've dated cousins of friends that have gotten me into a hell of a lot more trouble than they were ever worth.
