This video just came out a couple weeks ago: 17 minutes of mission footage where 47 has to escape an abandoned Chicago library while being hunted by Chicago's finest.

In short, this looks amazing. I was beginning to wonder what was taking them so long to release the next installment after Hitman: Blood Money. Now I see why. Looks like they pulled out all the stops.
The gameplay looks like a blend of Hitman, Arkham Asylum, Assassin's Creed, and Red Dead Redemption. No more maps, no saves, players can set elaborate traps, use instinct mode (similar to detective mode in Batman: AA), and can use a tag and kill feature similar to RDR. 47 also looks a lot more agile than he has in the past.
Immediate reactions to the video:
Pros: Glacier 2 engine looks awesome, much more immersive, cinematic dialogue, more "organic" feel to gameplay
Cons: David Bateson is not voicing Agent 47, Jesper Kyd is no longer helming the soundtrack (new one seems to overuse the "Hans Zimmer Effect" horn section), NO OFFICIAL RELEASE DATE YET (other than the vague 2012)!
