Leather is a natural non-homogenous material meaning the stiffness varies across its surface area. Naturally creases will start forming at areas with lower stiffness. In addition the presence of reinforcements like the toe cap, your walking style, the shape of your foot will influence creasing. In summary, creasing is a function of multiple variables.
This being said creasing can in some cases be a strong indicator for shoes being the wrong size, e.g. if a cap toes shoe breaks not behind the cap, but on the cap it is likely to be too short.

Minor fit issues I don't think it is likely. Considering the nature of natural materials. But major fit issues it should be possible. Things like Instep issues, too wide, too narrow, to short.
My EG boot is too narrow, so I have folds above and below the stress point across the stress point. Those folds will not exist if the shoes are properly width.
A loose shoe will also get more wrinkled because it won't be stretched as well over your feet.
A pair of shoes that is too long will have creases too far up the shoe - because where your feet flex when you walk is further up the shoes.
