Will the 2:1 ratio change the taste depending on the size of the drink / glass used or is that really negligible?

Shaking this drink over ice will add 1 to 2 oz. of water. If you keep the 2:1 proportion but increase the alcohol amount (say, 3 oz. of rum and 1.5 oz. of lime juice), and keep the amount of water added the same, you're going to end up with a drink that may be a little stronger than you'd like.

Needless to say different rums are different strengths. Shaking vs. stirring or room temperature will make a difference in the dilution. Different lime cultivars and degrees of ripeness produce different acidity and flavor. This is why the practice of hard and fast recipes is problematic... best to experiment and go with ones own judgment in any given culinary project.
